Deletion of papR was found to enhance adhesion of UTI89 to both bladder and kidney cell lines in a manner independent of type-1 fimbriae. We demonstrate PapR mediated post-transcriptional repression of the P-fimbriae phase regulator gene papI and postulate a role for such regulation in fimbrial cross-talk at the population level in UPEC.

The majority of Escherichia coli strains isolated from urinary tract infections have the potential to express multiple fimbriae. Two of the most common fimbrial adhesins are type 1 fimbriae and pyelonephritis-associated pili (Pap). Previous research has shown that induced, plasmid-based expression of a Pap regulator, papB, and its close homologues

The P fimbriae family of adhesins is epidemiologically associated with prostatitis, pyelonephritis (70-90% of strains), and sepsis. This family of adhesins is  P fimbriae (also known as pyelonephritis-associated pili) or P pili or Pap are chaperon-usher type (specifically of the π family) fimbrial appendages found on the  P-fimbriae have been found to be a virulence factor in both human and nonhuman primate urinary tract infections caused by Escherichia coli. 2014-01-01 1992-06-01 S fimbriae and type 1 fimbriae were equally common in resident and transient strains. The results indicate that not only P fimbriae, but also, certain capsules and the ability to produce the siderophore aerobactin might contribute to persistence of E. coli in the large intestine. Fimbrial (type 1, P, and S) and afimbrial adhesins, the unique virulence traits of uropathogenic Escherichia coli (UPEC), are well recognized for their role in the initial step of uropathogenesis. In this study, we investigated whether these adhesins are dispensable for UPEC in adherence and invasio … P fimbriae may facilitate invasion across the epithelial barrier into the bloodstream (Warren et al., 1988) and stimulate a mucosal inflammatory response (Hedges et al., 1992; Agace et al., 1993; Hedlund et al., 1996).

Results: In this study we investigate the association of P fimbriae proteins to OMVs from both genetically manipulated E. coli laboratory strains and a uropathogenic isolate. We found that the OMVs could carry functionally active P fimbriae proteins on their surface and we observed specific attachment of OMVs to beads coated with the P fimbrial galabiose receptor. P-fimbriae are heteropolymeric fibers carrying a Gal(al-4)Gal-specific PapG adhesin at its distal end. The pyelone-phritic strain DS17 expresses P-fimbriae from a single gene cluster. Fimbriae are either located at the poles of a cell or are evenly spread over its entire surface. Mutant bacteria that lack fimbriae cannot adhere to their usual target surfaces, and thus cannot cause diseases.

P fimbriae

P fimbriae are classically associated with pyelonephritis. While type 1 fimbriae recognise mannosylated receptors, the PapG adhesin of P fimbriae binds to Galα1-4Gal moieties in glycolipids of the host cell membrane and P fimbrial binding is therefore not inhibited by mannose. The receptors for E. coli P fimbriae are the globoseries of glycosphingolipids; Gal alpha 1-->4Gal beta-containing oligosaccharides bound to ceramide in the outer leaflet of the lipid bilayer.
